Single Cell Sequencing

OVERVIEW

The single cell sequencing shared resource provides automated single cell library preparation with the use of 10X Genomics Chromium iX and Connect devices. Automated single cell and nuclei barcoding and library preparation for 3’ GEX, 5’ GEX with feature bar coding, 5 GEX with VDJ BCR and/or VDJ TCR sequencing, ATAC sequencing and fixed tissue and cell single cell sequencing applications can be performed on 8 samples at one time. Multiplexing of samples can also performed. We are currently developing an automated data analysis pipeline as well.

Single Cell Sequencing Shared Resource Services

  • Faculty consultation and referral to Chromium Connect for technical advice
  • 3’ gene expression library preparation with quality control
  • Blastocyst microinjection of embryonic stem cells to create knockout mice.
  • 5’ gene expression library preparation with quality control
  • 5’ TCR single cell sequencing library preparation with quality control
  • 5’ BCR single cell sequencing library preparation with quality control
  • Single cell ATAC sequencing library preparation with quality control
  • 3” single cell library preparation from fixed tissue or cells and quality control
  • 3’ library preparation from previously prepared single cell cDNA
  • All service includes recommendation for submitting libraries for sequencing
  • Other technical consultation as needed (advice on sample preparation)


NOTE: Information from non-Moores Cancer Center principal investigators must be on file before utilizing Shared Resource facilities.
